I am trying to replace the rollers on a glass sliding door. I presume they are push in but I can't get them out.

Have you tried loosening that screw? Did this do anything?
I suspect the answer is to loosen that screw, then lever the roller carriage out with a small flathead screwdriver. You may come up against some resistance, but levering it out should work.
