Help please - I’m trying to replace handles and can’t get this screw out. My screwdrivers don’t hold in the grooves (which are rapidly loosing their shape).
i have basic tools only.
What can I try to get this out?

Considering you are looking to replace the handles, meaning you won't need to reuse the screws, you have a couple of options.
The first thing I would try is to use some pliers to grip the screw and rotate it out.
If this doesn't work, I would just cut the screw head off with a hacksaw, then pull the body of the screw through the door.

Ok next question - how do I get the screw out of the handle? It’s off the door now the heads are cut off my little pliers I have (and my little strength!) aren’t having any luck winding it out
Try spraying some WD40 into the bottom of the thread. This could help to loosen things up a bit.
After this, clamp the handle to something solid so that it doesn't move, then grip the screw with your pliers as low down as possible. You want the rotational force to be applied as close to the base as possible.
If you have no luck, maybe try some pliers with a longer handle. The longer the handle is, the more force you will be able to apply.
If this doesn't work, then you might have to cut the screw off and drill it out.
Give the first steps I have mentioned a try and let me know how you go. If we have to drill it out, I can provide further assistance.

If you are replacing with new handles why do you want to get the headless screw out of the old handle?

I just had another thought.
If you can't get them out with pliers, try cutting it down so there is around 5mm sticking out still, then using your hacksaw to cut a groove in the top of the shaft. You could then use a flathead screwdriver to rotate the shaft of the screw.
Just another option.

It's just a Sunday afternoon thought @Lou_L, but Bunnings Trojan used to make a large fencing plier with a toothed section proximal to the hinge.
With 9" handles - you can easily obtain a truly crushing hold on the bolt.
