I want to make a 2 metre six inch pvc fishing rod holder to place on my roof rack of my 4WD. It must have screw on ends at each end and have some sort of U mechanism to attach it to the roof rack.

Welcome to the Bunnings Workshop community. It's sensational to have you join us, and thanks for sharing your question about 6" PVC piping.
The equivalent of 6" is around 152.4mm, the pipe the store carries is 150mm which is very close. Below are the parts I suggest in order to create your fishing rod holder.
The threaded access coupling can be glued onto the pipe itself, and the front has a threaded fitting which the threaded access cap can fit on. The other end can be covered with a standard push on cap which also needs to be glued on. But if you wish to have end to end access you can install threaded fittings on both ends.
To anchor the pipe to your vehicle, I suggest looking at using the Airco galvanized strapping. Please note that your car should have a form of roof racking already attached. In the interest of health and safety, I suggest using a galvanized nut and bolt configuration to hold the strap in place. However, should you find a 150mm "U" pipe clamp that would be ideal as well.
Yes, you can use a galvanized nut and bolt on an aluminium pipe, but you must prevent direct metal-to-metal contact to avoid galvanic corrosion. Use a non-conductive barrier like a nylon washer, rubber sleeve to physically separate the steel from the aluminium.

Purchased from a recent garage sale. Four inch PVC. I intend storing steel rod in them - hopefully preventing rust. Both ends shown with the main cap which looks water-tight with an O-ring.

Can you still get those galvanised stepped brackets with nuts/washers and bent bolts for pipe? Something like this from memory?
Or make something similar.

The typical approach for connecting pipes to surfaces is to use saddle clips like this https://www.bunnings.com.au/kinetic-150mm-head-pipe-saddle-clips-2-pack_p4920108 . I imagine there will be a way to bolt it to your roof racks to hold it in place.
@Noyade that looks like these https://www.bunnings.com.au/antsig-antenna-mount-u-bolt_p4361347 , but unfortunately, they are too small for the 150mm pipe. It would be a good option, but unfortunately, I couldn't find anything large enough.

Thanks for asking this question, while Bunnings may not sell this, a lot of Electrical suppliers do, this item is called a conduit tube and
it has threaded ends and caps, they would also sell the mountings to attach this to your vehicle, depending on the model of your roof racks,
please bear in mind that these will be expensive, they are made for Tradies.
EricL, gave you a list of fiitings that could make something like this, however if you only have a rod or two, you should look at a 100mm system,
these are more common and if you nest your rods with the rods together, the eyelets won't catch on each other.
